中文摘要: 目的 基于超高效液相色谱-四极杆飞行时间质谱技术(UHPLC?TOF?MS)和网络药理学,探讨“易层”贴敷膏药治疗膝骨关节炎的作用机制。方法 采用UHPLC?TOF?MS对“易层”贴敷膏药化学成分进行定性分析。利用SwissTargetPrediction和GeneCards等生物信息数据库,搜集中药活性成分与膝骨关节炎疾病相关的分子靶标,将两者交集靶点进行筛选,获得核心靶点并构建蛋白相互作用网络,对交集靶点进行GO功能和KEGG通路富集分析。采用Cytoscape 3.9.1软件构建“化合物-靶点-通路”网络,预测“易层”贴敷膏药治疗膝骨关节炎的作用靶点。结果 共鉴定“易层”贴敷膏药化学成分216个,主要包含类别为黄酮类、萜类、香豆素类。靶蛋白相互作用分析筛选核心靶点 81 个,KEGG 富集分析揭示了“易层”贴敷膏药可能的作用机制,即通过调控 AMPK、PI3K?Akt、Toll样受体等信号传导途径对膝骨关节炎产生治疗作用。结论 “易层”贴敷膏药治疗膝骨关节炎可能依赖多成分、多靶点、多途径协同发挥其治疗作用。

Abstract:Objective To investigate the mechanism of action of “YiCeng” in the treatment of knee osteoarthritis(KOA)based on Ultra-high performance liquid chromatography combined with time-of-flight mass spectrometry(UHPLC-TOF-MS) and network pharmacology. Methods UHPLC-TOF-MS was used to qualitatively analyse the chemical constituents of“YiCeng”plaster. Bioinformatic databases such as SwissTargetPrediction and GeneCards were used to collect molecular targets related to KOA,and the intersection of the two was screened to obtain the key targets and construct the protein-protein interaction network,and the intersection targets were analysed for the enrichment of theGO function and KEGG pathway. Cytoscape 3.9.1 software was used to construct a “compound-target-pathway” network and predict the targets of “YiCeng” plaster for the treatment of KOA. Results A total of 216 chemical components were identified in the “YiCeng” plaster,mainly including flavonoids,terpenes,and coumarins. Target protein interaction analysis screened 81 key targets,and KEGG enrichment analysis revealed the possible mechanism of action of the “YiCeng” plaster,which was to treat KOA by regulating signaling pathways such as AMPK,PI3K-Akt,and Toll-like receptors. Conclusion The treatment of KOA with “YiCeng” plaster may depend on the synergistic effects of multiple components,targets and pathways.
