中文摘要: 核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体在类风湿性关节炎、骨关节炎及痛风性关节炎等疾病的发生发展中起着十分重要的作用。在关节炎性疾病中,NLRP3炎症小体表达上调,介导炎症反应,影响疾病进展。类风湿关节炎中,NLRP3炎症小体诱导促炎因子产生,调节辅助性T细胞比例,介导滑膜炎症;骨关节炎中,NLRP3介导促炎因子分泌,影响基质蛋白酶产生,诱导软骨细胞凋亡,促进滑膜炎症,加速关节退变;痛风性关节炎症中,巨噬细胞NLRP3炎症小体被尿酸单钠晶体激活,促进炎症反应。本文对NLRP3炎症小体激活机制及其在关节炎症疾病发病中的作用进行综述。
Abstract:The incidence rate of arthritis diseases including rheumatoid arthritis, osteoarthritis and gouty arthritis is increasing. Nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome plays an important role in the occurrence and development of various arthritis diseases. In arthritic diseases, the NLRP3 inflammasome is upregulated, mediates inflammatory responses, and affects disease progression. In rheumatoid arthritis, NLRP3 inflammasome induces the production of pro-inflammatory factors, regulates the proportion of helper T cells, and mediates synovial inflammation; in osteoarthritis, NLRP3 mediates the secretion of pro-inflammatory factors, affects the production of matrix proteases, and induces chondrocyte apoptosis to promote joint degeneration; in gouty joint inflammation, macrophage NLRP3 inflammasomes are activated by monosodium urate crystals (MSU) to promote inflammatory responses. This article reviews the mechanism of NLRP3 inflammasome activation and its role in the pathogenesis of joint inflammatory diseases.
