小檗碱通过抑制PI3K/Akt对2型糖尿病大鼠炎症因子水平和糖脂代谢的影响

中文摘要: 目的 研究小檗碱基于磷脂酰肌醇3激酶(PI3K)/蛋白激酶B(Akt)通路对2型糖尿病(T2DM)大鼠炎症水平和糖脂代谢的影响。 方法选取40只SPF级SD雄性大鼠,其中随机选取10只实验大鼠为正常组,其余30只建造T2DM模型[随机分为模型组(10只)、二甲双胍阳性对照组(10只)、小檗碱组(10只)]。检测大鼠血清炎症因子[肿瘤坏死因子-α(TNF-α)、 C-反应蛋白(CRP)、白细胞介素-1β(IL-1β)],脂代谢指标[低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、三酰甘油(TG)、总胆固醇(TC)、游离脂肪酸(FFA)]和糖代谢指标[空腹胰岛素(FINS)、空腹血糖(FBG)、胰岛素抵抗指数(HOMA-IR)],检测胰腺组织中PI3K、Akt和p-PI3Kp85蛋白的表达。 结果 T2DM模型三个组的血清糖代谢指标(FBG、FINS、HOMA-IR)、脂代谢指标(TC、TG、LDL-C、FFA)、炎症因子(TNF-α、CRP、IL-1β)水平均高于正常组(P<0.05);T2DM模型三个组中,依模型组、二甲双胍阳性对照组、小檗碱组之序, 除HDL-C水平递升,其他指标水平均递降(P<0.05)。T2DM模型三个组胰腺组织中的PI3K、Akt和p-PI3Kp85蛋白相对表达水平均低于正常组(P<0.05),且三者表达水平依模型组、二甲双胍阳性对照组、小檗碱组之序递升(P<0.05)。 结论 小檗碱能改善T2DM大鼠脂代谢紊乱状况,可抑制其体内炎症反应,作用机制可能与调控PI3K/Akt通路有关。
Abstract:Objective To study the effects of berberine on inflammatory factor levels and glucose and lipid metabolism in type 2 diabetes mellitus (T2DM) rats based on phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt) pathway. Methods Forty SPF grade SD male rats were selected, of which 10 rats were randomly selected as normal group, and the other 30 rats were constructed as T2DM model rats [divided into model group (10 rats), metformin positive control group (10 rats) and berberine group (10 rats) randomly]. The levels of inflammatory factors [tumor necrosis factor-α (TNF-α)、 C-reactive protein (CRP), interleukin-1β (IL-1β)], lipid metabolism indexes [low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), triglyceride (TG), total cholesterol (TC), free fatty acid (FFA)], and glucose metabolism indexes [fasting insulin (FINS), fasting blood glucose (FBG), insulin resistance index (HOMA-IR)]. The expressions of PI3K, Akt and p-PI3Kp85 proteins in pancreatic tissue were detected. Results The levels of serum glucose metabolism indexes (FBG, FINS, HOMA-IR), lipid metabolism indexes (TC, TG, LDL-C, FFA) and inflammatory factors (TNF-α, CRP, IL-1β) in the three T2DM model groups were higher than those in the normal group (P<0.05). Among the three T2DM model groups, except for HDL-C levels that gradually increased, all other indicator levels decreased gradually in the order of model group, metformin positive control group and berberine group (P<0.05). The relative expression levels of PI3K, Akt and p-PI3Kp85 proteins in pancreatic tissues of the three T2DM model groups were lower than those of the normal group (P<0.05), while the relative expression levels of the three proteins increased sequentially in order of model group, metformin-positive control group and berberine group (P<0.05). Conclusion Berberine can improve the disorder of lipid metabolism in T2DM rats and inhibit the inflammatory reaction in vivo. The mechanism may be related to the regulation of PI3K/Akt pathway.
