输血对脓毒症大鼠急性肺损伤肺上皮细胞活性、肺超微结构及SIRT1/PGC-1α的影响

中文摘要: 目的 探讨输血对脓毒症大鼠急性肺损伤肺上皮细胞活性、肺超微结构及沉默信息调节子1(SIRT1)/过氧化物酶体增殖物激活受体γ共激活因子-1α(PGC-1α)蛋白表达的影响。 方法 从50只SPF级SD雄性大鼠中选取35只建模,建模成功后随机选取30只,分为模型(B)组,液体复苏(C)组,输血(D)组,每组10只;从剩余的15只中随机选取10只作为正常对照(A组)。对C组实施液体复苏治疗,对D组实施输血治疗,A组、B组不给药,记录肺系数并用干湿重法测定肺含水量(LWC),HE染色法检测肺组织病理形态,电子显微镜观察肺超微结构,TUNEL法检测肺上皮细胞活性,免疫印迹法检测大鼠肺组织中SIRT1/PGC-1α蛋白表达。 结果 与A组比较,B组肺系数、LWC、肺上皮细胞凋亡率明显升高(P<0.05),肺组织SIRT1/PGC-1α蛋白表达显著降低(P<0.05),与B组比较,C组、D组的肺系数、LWC、肺上皮细胞凋亡率明显降低(P<0.05),肺组织SIRT1/PGC-1α蛋白表达显著升高(P<0.05),且D组比C组变化明显(P<0.05);A组肺组织结构清晰完整,未见明显病理改变,B组可见明显病理改变,与B组比较,C组、D组病理症状明显改善,且D组改善更为明显;A组肺组织超微结构正常,B组肺组织超微结构出现明显异常,与B组相比,C组、D组肺组织超微结构明显改善,且D组改善最为明显。 结论 输血对脓毒症大鼠有很好的疗效,可显著改善肺损伤、肺上皮细胞活性及肺超微结构,并促进SIRT1/PGC-1α蛋白表达。
Abstract:Objective To investigate the effects of blood transfusion on lung epithelial cell activity, lung ultrastructure and Silent information regulator 1(SIRT1)/Peroxisome proliferator-activated receptor γ coactivator-1α(PGC-1α) protein in septic rats with acute lung injury. Methods Thirty rats were selected from 50 male SPF grade SD rats for modeling, after successful modeling, 30 rats were randomly selected and divided into model (B) group, fluid resuscitation (C) group, and blood transfusion (D) group, with 10 rats in each group; Ten of the remaining 15 rats were randomly selected as normal controls (group A). Group C was given liquid resuscitation treatment, and group D was given blood transfusion treatment, while group A and B were not given drugs. The lung coefficient was recorded and the lung water content(LWC) was measured by dry and wet weight method. The pathological morphology of lung tissue was detected by HE staining, the ultrastructure of lung was observed by electron microscope, the activity of lung epithelial cells was detected by TUNEL method, and the SIRT1/PGC-1α protein expression in rat lung tissue was detected by Western Blot. Results Compared with group A, the lung coefficient and LWC and apoptosis rate of lung epithelial cells were significantly increased in group B(P<0.05), and the expression of SIRT1/PGC-1α protein in lung tissue was significantly decreased(P<0.05). Compared with group B, the lung coefficient, LWC and apoptosis rate of lung epithelial cells were significantly decreased(P<0.05), and the expression of SIRT1/PGC-1α protein in lung tissue was significantly increased in groups C and D(P<0.05), while the change of group D was significantly higher than those of group C(P<0.05). The lung tissue structure of group A was clear and complete with no obvious pathological changes, while obvious pathological changes were observed in group B. Compared with group B, the pathological symptoms of group C and D were significantly improved, and the improvement was more obvious in group D. The ultrastructure of lung tissue in group A was normal, while that of lung tissue in group B was obviously abnormal. Compared with group B, the ultrastructure of lung tissue in groups C and D was significantly improved, and the improvement in group D was the most obvious. Conclusion Blood transfusion has a good effect on sepsis rats, and can significantly improve lung injury, lung epithelial cell activity and lung ultrastructure, and promote the expression of SIRT1/PGC-1α protein.
