中文摘要: 目的 探讨过表达整合素β6对人结肠癌SW480细胞基因表达的影响,揭示整合素β6促进人结肠癌细胞侵袭、转移的潜在机制。方法通过慢病毒载体构建稳定过表达整合素β6的SW480细胞株(SW480/β6-OE)和空载体转染的SW480细胞株(SW480/Mock),western blot检测整合素β6过表达效率,Transwell实验检测过表达整合素β6对SW480细胞侵袭能力的影响。高通量转录组测序技术分析过表达整合素β6后SW480细胞基因表达谱的变化,并对差异表达基因(DEGs)进行GO和 KEGG功能富集分析。结果 过表达整合素β6显著增强SW480细胞的侵袭能力(P<0.05)。相对于SW480/Mock细胞,在SW480/β6-OE细胞中共筛选出1 472个DEGs,其中上调基因840个,下调基因632个。KEGG富集分析显示,DEGs显著富集于癌症通路、MAPK信号通路、PI3K-AKT信号通路、细胞因子与其受体互作、cAMP信号通路和Wnt信号通路等。结论 过表达整合素β6促进结肠癌细胞的侵袭、转移,改变结肠癌细胞的基因表达谱,DEGs 显著富集的信号通路包括多条已被证实在结肠癌的发生发展中发挥重要作用的信号通路。
Abstract:Objective To explore the effects of overexpression of integrin β6 on gene expression of human colon cancer SW480 cells to reveal a potential mechanism of integrin β6 in promoting invasion and metastasis of human colon cancer cells. Methods SW480 cell line stably overexpressing integrin β6 (SW480/β6-OE) and SW480 cells transfected with empty vector (SW480/Mock) were established by lentivirus vector. The overexpression efficacy of integrin β6 was detected by western blot, and the effect of overexpression integrin β6 on the invasion ability of SW480 cells was detected by transwell assay. High throughput transcriptome sequencing technology was used to analyze the changes in gene expression profile of SW480 cells after overexpression of integrin β6, and the GO and KEGG function enrichment analysis was conducted for differentially expressed genes(DEGs). Results Overexpression of integrin β6 significantly enhanced the invasion ability of SW480 cells(P<0.05). Compared with SW480/Mock cells, 1472 DEGs were screened from SW480/β6-OE cells, including 840 up-regulated genes and 632 down-regulated genes. KEGG enrichment analysis revealed that DEGs were enriched in the pathways involved in cancer, mitogen-activated protein kinase (MAPK), PI3K-AKT, cytokine-cytokine receptor interaction, cAMP signal pathway and Wnt signal pathway. Conclusions Overexpression of integrin β6 promotes the invasion and metastasis of SW480 cells and changes the gene expression profile of colon cancer cells. The multiple signaling pathways in which DEGs are significantly enriched have been confirmed to play an important role in the occurrence and development of colon cancer.
