miRNA-140-5p低表达促进人骨髓间充质干细胞的成骨分化

中文摘要: 目的 探讨miRNA-140-5p对人骨髓间充质干细胞(hBMSCs)成骨分化的影响及其可能的相关机制。方法 构建miRNA-140-5p模拟物(Mimic组)、抑制剂(Inhibitor组)、重组质粒pcDNA3.1-Wnt1及其相应的阴性对照并转染hBMSCs,应用CCK-8、茜素红染色、Elisa、qRT-PCR、Western blot等多种分子生物学技术检测hBMSCs的细胞活力和成骨分化情况,应用双荧光素酶报告基因系统验证miRNA-140-5p的靶基因。结果 转染后,Mimic组miRNA-140-5p表达水平显著升高,Inhibitor组显著降低 (P<0.05),而hBMSCs的细胞活力在两组均无明显变化(P>0.05)。茜素红染色和成骨分化标志物Runt相关转录因子2(RUNX2)、碱性磷酸酶(ALP)蛋白检测结果显示Inhibitor组hBMSCs的成骨分化明显升高 (P<0.05),Mimic组hBMSCs的成骨分化受到抑制 (P<0.05)。双荧光素酶报告基因系统结果显示,Wnt1是miRNA-140-5p的靶基因。Western blot检测显示Wnt1和β-联蛋白(β-catenin)的表达水平在Mimic组降低 (P<0.05),在Inhibitor组升高 (P<0.05)。在hBMSCs中高表达Wnt1后,其成骨分化明显升高 (P<0.05)。结论 miRNA-140-5p的低表达可以通过靶向Wnt1/β-catenin信号通路促进hBMSCs的成骨分化。
Abstract:Objective To investigate the effect of miRNA-140-5p on the osteogenic differentiation of human bone marrow mesenchymal stem cells (hBMSCs) and its possible mechanisms. Methods The miRNA-140-5p mimic, inhibitor, recombinant plasmid pcDNA3.1-Wnt1 and its corresponding negative control were constructed and transfected into hBMSCs,which were divided into corresponding groups according to the different transfectants. CCK8, alizarin red staining, ELISA, qRT-PCR, Western blot and other molecular biological techniques were applied to detect the cell viability and osteogenic differentiation of hBMSCs, and the dual-luciferase reporter gene system was used to identify the target gene of miRNA-140-5p. Results After transfection, the expression level of miRNA-140-5p was significantly increased in Mimic group and was significantly decreased in Inhibitor group(P<0.05), while the cell viability of hBMSCs had no significant change in both groups(P>0.05). Alizarin red staining and the detection of osteogenic differentiation markers Runt-related transcription factor 2(RUNX2) and alkaline phosphatase (ALP) protein showed that the osteogenic differentiation of hBMSCs was significantly increased in Inhibitor group and was inhibited in Mimic group(P<0.05). The results of the dual-luciferase reporter gene system showed that Wnt1 was the target gene of miRNA-140-5p. Western blot analysis showed that the expression levels of Wnt1 and β-catenin decreased in Mimic group and increased in Inhibitor group(P<0.05). High expression of Wnt1 in hBMSCs significantly increased their osteogenic differentiation(P<0.05). Conclusion Low expression of miRNA-140-5p can promote the osteogenic differentiation of hBMSCs by targeting the Wnt1/β-catenin signaling pathway.
