中文摘要: 目的 探讨猫白血病病毒C亚类受体1反义RNA1(FLVCR1-AS1)对宫颈癌细胞SiHa增殖、迁移和侵袭的影响及可能机制。方法 收集2016年12月至2018年11月在海南医学院第二附属医院行手术治疗的35例宫颈癌患者的癌组织和癌旁组织。体外培养SiHa细胞,按不同转染分为si-NC组、小干扰RNA(si)-FLVCR1-AS1组、miR-NC组、微小RNA(miR)-877-5p组、si-FLVCR1-AS1+anti-miR-NC组、si-FLVCR1-AS1+anti-miR-877-5p组,CCK-8法检测细胞增殖,Transwell检测细胞迁移和侵袭,Western Blot法检测细胞中Ki-67、基质金属蛋白酶(MMP)-2和MMP-9蛋白表达,RT-qPCR检测FLVCR1-AS1和miR-877-5p表达。双荧光素酶报告基因实验验证FLVCR1-AS1和miR-877-5p的调控关系。结果 宫颈癌组织中FLVCR1-AS1表达量高于癌旁组织(3.82±0.23 vs 1.00±0.08,t=68.510, P<0.01);而miR-877-5p表达量低于癌旁组织(0.32±0.03 vs 1.00±0.05,t=68.993, P<0.01)。经转染后,si-FLVCR1-AS1组SiHa细胞中FLVCR1-AS1表达较si-NC组显著下调(P<0.01),且其OD值、迁移数、侵袭数及Ki-67、MMP-2和MMP-9蛋白的表达量均低于si-NC组(P<0.01)。miR-877-5p组SiHa细胞中miR-877-5p表达较miR-NC组显著上调(P<0.01),且其OD值、迁移数、侵袭数及Ki-67、MMP-2和MMP-9蛋白表达量均低于miR-NC组(P<0.01)。与miR-877-5p抑制剂共转染后,si-FLVCR1-AS1+anti-miR-877-5p组SiHa细胞OD值、迁移数、侵袭数及Ki-67、MMP-2和MMP-9蛋白表达量均高于si-FLVCR1-AS1+anti-miR-NC组(P<0.01)。结论 FLVCR1-AS1可能通过靶向抑制miR-877-5p的表达促进宫颈癌细胞SiHa增殖、迁移和侵袭。
Abstract:Objective To investigate the effect of feline leukemia virus subgroup C receptor 1-antisense 1 (FLVCR1-AS1) on the proliferation, migration and invasion of SiHa cervical cancer cells and its possible mechanism. Methods The cancer tissues and adjacent tissues were collected from 35 patients with cervical cancer who underwent surgery in the Second Affiliated Hospital of Hainan Medical College from December 2016 to November 2018. SiHa cells were cultured in vitro and transfected respectively with negative control siRNA (si-NC group), si-FLVCR1-AS1(si-FLVCR1-AS1 group), miR-NC(miR-NC group),miR-877-5p mimics(miR-877-5p group)and co-transfected with si-FLVCR1-AS1 and anti-miR-NC(si-FLVCR1-AS1+anti-miR-NC group)and with si-FLVCR1-AS1 and anti-miR-877-5p(si-FLVCR1-AS1+anti-miR-877-5p group). CCK-8 method was used to detect the cell proliferation, and transwell method was used to detect the migration and invasion of cells. The protein expressions of Ki-67, MMP-2 and MMP-9 were determined by western blot method and the expressions of FLVCR1-AS1 and miR-877-5p were detected by RT-qPCR. The regulatory relationship between FLVCR1-AS1 and miR-877-5p was confirmed by double-luciferase reporter gene assay. Results The expression level of FLVCR1-AS1 in cervical cancer tissue was significantly higher than that in adjacent tissue (3.82±0.23 vs 1.00±0.08,t=68.510, P<0.01), and the expression level of miR-877-5p was lower than that in adjacent tissue (0.32±0.03 vs 1.00±0.05,t=68.993,P<0.01). After transfection, the expression of FLVCR1-AS1 in SiHa cells was significantly down-regulated in si-FLVCR1-AS1 group compared to si-NC group (P<0.01),the expression of miR-877-5p in SiHa cells was significantly up-regulated in miR-877-5p group compared to miR-NC group (P<0.01), and their OD value, migration and invasion of SiHa cells and the protein expressions of Ki-67, MMP-2 and MMP-9 in si-FLVCR1-AS1 group and in miR-877-5p group were significantly lower than those in si-NC group and in miR-NC group, respectively (P<0.05). After co-transfection with miR-877-5p inhibitor, the OD value, migration number, invasion number and protein expression of Ki-67, MMP-2 and MMP-9 of SiHa cells in si-FLVCR1-AS1+anti-miR-877-5p group were higher than those in si-FLVCR1-AS1+anti-miR-NC group. Conclusion FLVCR1-AS1 may promote the proliferation, migration and invasion of SiHa cervical cancer cells by targeted inhibition of miR-877-5p expression.
