子宫内膜癌中脂肪酸代谢重编程的关键酶研究进展

中文摘要: 子宫内膜癌是女性生殖系统三大恶性肿瘤之一,发病率及死亡率在全球范围内逐年升高。早期子宫内膜癌患者预后良好,但晚期及复发性子宫内膜癌的5年生存率不足40%。近年来,免疫和分子靶向治疗与传统化疗的联合应用使该部分患者生存获益明显,但当单独应用靶向药物或免疫检查点抑制剂时,肿瘤的客观缓解率未得到显著改善。因此,寻求更为高效的治疗靶点对治疗晚期及复发性子宫内膜癌至关重要。肿瘤代谢重编程因其可改变肿瘤细胞代谢模式满足细胞增殖和生长的能量需求而逐渐受到关注,既往子宫内膜癌的代谢改变研究多集中于糖代谢,而现有研究表明脂肪酸代谢变化亦在其中发挥重要作用。脂肪酸代谢重编程包括脂肪酸合成、摄取、以及氧化分解的失衡,代谢过程中催化相关反应的酶蛋白关键因子的异常表达是脂肪酸代谢失衡的关键。因此探究子宫内膜癌脂肪酸代谢改变及脂肪酸代谢相关酶蛋白等关键因子在脂肪酸代谢通路中的作用和变化,对寻找子宫内膜癌新的治疗方法和潜在高效治疗靶点起着重要的作用。
Abstract:Endometrial cancer is one of the three major malignant tumors in the female reproductive system, with the incidence and mortality rates increasing globally. Early-stage endometrial cancer patients have a good prognosis, but the 5-year survival rate for advanced and recurrent endometrial cancer is less than 40%. In recent years, the combined use of immunotherapy and molecular targeted therapy with traditional chemotherapy has significantly improved the survival of these patients. However, when targeted drugs or immune checkpoint inhibitors are used alone, the objective response rate of the tumors is not significantly improved. Therefore, it is crucial to seek more efficient treatment targets for advanced and recurrent endometrial cancer. Tumor metabolism reprogramming has attracted attention due to its ability to change the metabolic patterns of tumor cells to meet their energy needs for proliferation and growth. Previous studies on metabolic changes in endometrial cancer have mainly focused on carbohydrate metabolism, but existing research suggests that changes in fatty acid metabolism also play an important role. Fatty acid metabolism reprogramming includes imbalances in fatty acid synthesis, uptake, and oxidation. Abnormal expression of enzyme protein key factors catalyzing related reactions in the metabolic process is the key to the imbalance of fatty acid metabolism. Therefore, exploring the changes in fatty acid metabolism and the role and changes of key factors such as fatty acid metabolism-related enzyme proteins in the fatty acid metabolism pathway plays an important role in finding new treatment methods and potential efficient treatment targets for endometrial cancer.
