中文摘要: 血管生成拟态(VM)是由肿瘤细胞及细胞外基质共同形成的类似血管的结构,这种结构可以模仿血管的功能并为肿瘤提供营养。肿瘤血管的生成在肿瘤细胞生长、侵袭和转移中起重要作用,抗血管生成药物已应用于乳腺癌治疗中,但药物作用尚具有局限性且易发生耐药,VM的形成是导致抗血管生成药物耐药的原因之一。虽然VM的形成机制尚未完全明确,但是在乳腺癌中其与多种预后不良因素相关,如更具侵袭性的分型、更大的肿瘤直径及更易发生淋巴结转移等,早期干预VM的形成或可改善乳腺癌预后,因此靶向VM的治疗可能成为乳腺癌治疗的一大新方向。本文主要针对VM形成的分子机制、乳腺癌中相关VM的驱动因素、VM在乳腺癌中的临床相关性及有关靶向VM的治疗进行综述。
Abstract:Vasculogenic mimicry (VM) is a blood vessel-like structure formed by tumor cells and extracellular matrix, which can mimic the function of blood vessels while providing nutrients to tumors. Tumor angiogenesis plays an important role in tumor cell growth, invasion and metastasis. Anti-angiogenic drugs have been applied in breast cancer treatment, but the drug effects are still limited and susceptible to drug resistance. The formation of VM is one of the reasons leading to resistance to anti-angiogenic drugs. In addition, although the mechanism of VM formation has not been completely clarified, it is associated with a variety of poor prognostic factors in breast cancer, such as more aggressive staging, larger tumor diameter, and more prone to lymph node metastasis. Early intervention in the formation of VM may improve the prognosis of breast cancer, and therefore, targeting the VM may become a major new direction in the treatment of breast cancer. This article focuses on the molecular mechanisms of VM formation, the drivers of associated VM in breast cancer, the clinical relevance of VM in breast cancer, and the treatment targeting at VM.
