中文摘要: 目的 研究在三阴性乳腺癌(TNBC)中上皮—间质转化标志物——波形蛋白(Vimentin)的表达及其与乳腺癌特异性基因1(BCSG1)的关系。方法 选取2018年1月至2020年1月于长沙市第三医院接受治疗的女性TNBC患者60例作为研究对象,另选取同期接受治疗的乳腺良性病变患者20例作为对照组。运用qRT-PCR、免疫组化法检测Vimentin、BCSG1在TNBC组织与乳腺良性病变组织中的表达水平。结果 Vimentin和BCSG1表达水平在乳腺良性病变组织(1.02±0.05,0)、non-TNB组织(2.13±0.21,2.16±0.23)和TNBC组织(2.41±0.22,2.45±0.24)中依次增高(F=382.600,P<0.01;F=1 003.000,P<0.01)。TNBC中的Vimentin与BCSG1表达具有正相关关系(r=0.370, P<0.05),Vimentin、BCSG1高表达是影响TNBC患者预后的独立危险因素(OR=13.662, P=0.035; OR=15.443, P=0.001)。Vimentin联合BCSG1对TNBC诊断的灵敏度为92.35%,特异度为95.64%,ROC曲线下面积为0.891。结论 BCSG1与Vimentin具有正相关关系,其在TNBC组织中的表达水平均明显高于乳腺良性病变组织,二者联合对TNBC有较高诊断效能。
Abstract:Objective To explore the expression of Vimentin, a marker of epithelial-mesenchymal transition, and its association with breast cancer specific gene 1(BCSG1) in triple negative breast cancer(TNBC).
Methods Sixty female patients with TNBC who received treatment in the Third Hospital of Changsha from January 2018 to January 2020 were selected as the research subjects(research group), and 20 female patients with benign breast lesions receiving treatment at the same period were served as control group. Vimentin and BCSG1 expression in TNBC cancerous tissues and benign breast lesions were detected by qRT-PCR and immunohistochemistry.
Results Vimentin and BCSG1 expression levels increased sequentially in benign breast lesion tissues (1.02±0.05, 0), non-TNB tissues (2.13±0.21, 2.16±0.23) and TNBC tissues (2.41±0.22, 2.45±0.24) (F=382.600, P<0.01; F=1003.000, P<0.01). There was a positive correlation between the expression of Vimentin and BCSG1 in TNBC (r=0.370, P<0.05), and the high expressions of Vimentin and BCSG1 were the independent risk factors for the prognosis of TNBC patients (OR=13.662, P=0.035; OR=15.443, P=0.001). The sensitivity of Vimentin combined with BCSG1 for TNBC diagnosis was 92.35%, the specificity was 95.64%, and the area under the ROC was 0.891.
Conclusion BCSG1 and Vimentin have a positive correlation, and their expression levels in TNBC tissues are significantly higher compared with benign breast lesions, and the combination of the two has high diagnostic efficacy for TNBC.
