干扰素α-2b抑制大鼠术后肠粘连的实验研究

中文摘要: 目的探讨干扰素α-2b对大鼠术后肠粘连的抑制作用。方法选取60只SD大鼠,雌雄各半,随机分成6组,依次为正常对照组、生理盐水组、4个实验组(A组、B组、C组、D组),每组 10 只大鼠。除正常对照组外,其余5组均行锉刀摩擦盲肠术方法建立肠粘连模型。术后次日起,每日进行腹腔注射,生理盐水组注射生理盐水10 ml/kg,实验A、B、C、D组分别注射不同浓度(浓度依次为1×106 u/L、2×106 u/L、4×106 u/L、8×106 u/L)的干扰素α-2b 10 ml/kg,每日1次,连续注射7 d。6组均于术后第8日开腹,观察肠粘连程度并进行Nair分级,取相应部位的盲肠组织进行HE染色切片,观察病理改变并测定羟脯氨酸含量。结果(1)肠粘连程度:与正常对照组相比,其余5组肠粘连发生率及粘连严重级别均显著增高(P<0.05);与生理盐水组相比,实验A组、B组、C组无明显改变(P>0.05),D组肠粘连发生率有所下降,粘连严重级别显著降低(P<0.05)。(2)羟脯氨酸含量:与正常对照组相比,其余5组羟脯氨酸含量均明显增高(P<0.05);与生理盐水组相比,实验A组、B组则无明显变化(P>0.05),C组、D组羟脯氨酸含量明显降低(P<0.05);D组较C组更低(P<0.05)。(3)病理改变:与正常对照组相比,其余5组大多有大量的炎性细胞和成纤维细胞浸润;与生理盐水组相比,A组、B组无明显变化,C组、D组炎性细胞和成纤维细胞浸润明显减少。结论在一定浓度下,干扰素α-2b可以抑制成纤维细胞增殖以及胶原蛋白合成,术后注射可明显降低大鼠肠粘连的发生。
Abstract:Objective To explore the effect of interferon α-2b on inhibiting postoperative intestinal adhesion in rats. Methods Sixty SD rats(half male and half female) were randomly divided into 6 groups:normal control group,normal saline group and experimental groups (group A,group B,group C,group D),with 10 rats in each group.Except for the normal control group,the other 5 groups were treated with file friction cecum to establish the model of intestinal adhesion.From the next day after operation,intraperitoneal injection was performed.The rats in the normal saline group were injected with normal saline 10 ml/kg,and the rats in the experimental groups A,B,C and D were injected with different concentrations (1×106 u/L,2×106 u/L,4×106 u/L,8×106 u/L)of interferon α-2b 10 ml/kg,once a day for 7 days.The rats in six groups underwent laparotomy on the 8th day after operation to observe the degree of intestinal adhesion and perform Nair grade.The corresponding cecal tissues were taken for HE staining section to observe the pathological changes and determine the content of hydroxyproline. Results (1) Degree of intestinal adhesion:compared with the normal control group,the incidence and severity of intestinal adhesion in the other five groups were significantly higher (P<0.05).Compared with the normal saline group,there were no significant changes in group A,group B and group C (P>0.05),while the incidence of intestinal adhesion decreased,and the severity of adhesion decreased significantly in group D(P<0.05).(2) Hydroxyproline content:compared with the normal control group,the hydroxyproline content of the other five groups were significantly increased (P<0.05).Compared with the normal saline group,there were no significant changes in group A and group B (P>0.05),but the content of hydroxyproline significantly reduced in group C and group D (P<0.05),and that in group D was lower than in group C (P<0.05).(3)Pathological changes:compared with the normal control group,most of the other five groups had a large number of inflammatory cells and fibroblasts infiltration.Compared with the normal saline group,there were no significant changes in group A and group B,while the infiltration of inflammatory cells and fibroblasts was significantly reduced in group C and group D. Conclusion At a certain concentration,interferon α-2b can inhibit fibroblast proliferation and collagen synthesis,and postoperative injection of interferon α-2b can significantly reduce the occurrence of intestinal adhesions in rats.
