中文摘要: 目的 构建E3泛素化连接酶环指与WD重复结构域蛋白(RING finger and WD repeat domain,RFWD3)的慢病毒表达载体,研究RFWD3对宫颈癌细胞增殖与迁移能力的影响。方法 以宫颈癌HeLa细胞的cDNA为模板扩增RFWD3基因片段,利用DNA重组技术,将上述片段克隆至慢病毒载体pCDH-CMV-MCS-EF1-copGFP(简称pCDH)中,构建pCDH-RFWD3重组真核表达质粒。经酶切及测序验证准确后,采用三质粒慢病毒包装系统包装RFWD3病毒,通过梯度稀释法感染人胚肾上皮细胞(293T)检测病毒滴度。随后采用RFWD3慢病毒液感染宫颈癌HeLa细胞,运用Western blot实验检测慢病毒转导后HeLa细胞中RFWD3蛋白的表达水平,进一步通过CCK-8和细胞划痕实验检测过表达RFWD3蛋白对HeLa细胞增殖与迁移能力的影响。结果 可表达RFWD3蛋白的pCDH-RFWD3慢病毒载体获成功构建。包装RFWD3慢病毒,建立稳定表达RFWD3蛋白的宫颈癌HeLa细胞系。RFWD3慢病毒转导后的HeLa细胞与对照细胞相比,其细胞增殖与迁移能力显著增强(P<0.05)。结论 RFWD3能够显著增强宫颈癌HeLa细胞的增殖与迁移能力。
Abstract:ObjectiveTo investigate the effects of RING finger and WD repeat domain(RFWD3)on cervical cancer cells proliferation and migration by constructing the recombinant lentivirus vector which carries RFWD3.MethodsThe gene fragment of RFWD3 protein was amplified with the cDNA of HeLa cells,and was cloned into the lentiviral vector pCDH-CMV-MCS-EF1-copGFP (pCDH) plasmid by DNA recombination technique to construct the recombinant eukaryotic expression-vector of pCDH-RFWD3.The plasmid was verified to be corrected by enzymatic digestion and sequencing,and then RFWD3 fusion virus was obtained by lentivirus packaging method,and infected human embryonic kidney epithelial cells (293T) by gradient dilution method to detect the virus titer.Western blot assay was used to detect the expression of RFWD3 protein in HeLa cells after RFWD3 fusion virus infection,and the effect of up-regulated RFWD3 expression on the proliferation and migration ability of HeLa cells was studied by CCK-8 method and cell scratch test.ResultsThe pCDH-RFWD3 plasmid carrying RFWD3 gene was successfully constructed.Three plasmids were co-transfected into Human Embryonic Renal Epithelial cells (293T) by lentivirus packaging to construct RFWD3 gene fusion lentivirus.The level of RFWD3 protein was significantly increased and the cell proliferation and migration activity were significantly increased in cervical cancer cells which infected with lentivirus(P<0.05).ConclusionStudies have demonstrated that RFWD3 can significantly promote the proliferation and migration ability of HeLa cells.
