Abstract:Objective To analyze the medication patterns used by Professor ZHANG Xingping in treating insomnia of heart not storing spirit using data mining methods, and to explore the mechanisms of action of the core prescriptions through network pharmacology techniques.Methods Established a database for Professor ZHANG Xingpings treatment of insomnia of heart not storing spirit on the Traditional Chinese Medicine Inheritance Computational Platform (V3.0), compiled statistics on drug usage frequency, characteristics, and meridian attribution, and conducted association rule analysis and cluster analysis to mine core drug combinations. Collected target sites of the core prescriptions and those associated with insomnia of heart not storing spirit, screened common targets, constructed a protein-protein interaction (PPI) network to identify key targets, and analyzed the mechanisms of action using enrichment analysis methods. Results A total of 288 prescriptions were included, containing 145 types of Chinese materia medica, with 12 drugs having a usage frequency greater than 200 times. The nature of the medications was predominantly cold, the flavors primarily bitter and pungent, and the channel tropism mainly to the liver and spleen meridians; the primary efficacy was to clear heat, nourish deficiency, and regulate Qi. Analysis of the formulating rules yielded a core prescription including Huanglian, Huangqin, Xiakucao, Chaihu, Chishao, Baishao, Yujin, Chenpi, Banxia, Baizhu, Fuling, and Gancao. The core targets for treating insomnia of heart not storing spirit with the core prescription were AKT1, IL-1β， IL-6, CASP3, etc.； the biological processes involved primarily include responses to lipopolysaccharides, regulation of blood vessel diameter, vascular processes in the circulatory system, responses to oxidative stress, etc.; the main pathways involved include lipids and arteriosclerosis, TNF pathway, IL-17 pathway, etc., involving mechanisms such as inflammation and apoptosis. Conclusion Professor ZHANG Xingpings treatment of insomnia of heart not storing spirit focuses on the heart while also considering the liver and spleen, adopting a method of clearing the heart and calming the spirit, supplemented by soothing the liver and securing the soul, and moving the spleen to preserve intention. The mechanism of action of the core prescription, Qingxin Ningshen Decoction, in treating insomnia of heart not storing spirit may be related to regulating inflammatory responses and cellular apoptosis.
