Abstract:Objective To explore the expression levels of inflammasome absent in melanoma 2(AIM2) and gasdermin-D(GSDMD) in cervical squamous cell carcinoma and their relationship with clinicopathological features. Methods The expression levels of AIM2 inflammasome and GSDMD in cervical squamous cell carcinoma tissues were observed using the Cancer Genome Atlas (TCGA) database. From June 2019 to June 2021, 80 cases of cancer tissue samples of cervical squamous cell carcinoma patients (case group) and 45 cases of normal cervical tissue samples of uterine fibroid (control group) which obtained by surgery in Xuzhou Central Hospital were collected to detect the expression levels of AIM2 inflammasome and GSDMD by immunohistochemistry(IHC) and polymerase chain reaction (PCR). The associations of AIM2 inflammasome and GSDMD levels with the clinicopathological characteristics were analyzed. Results TCGA database revealed that compared with normal cervical tissues, the expression level of AIM2 inflammasome was sinificantly increased (P＜0.01), while GSDMD was mildly increased in carcinoma tissues (P＞0.05). Both IHC and PCR showed that the expression levels of AIM2 inflammasome and GSDMD in case group were significantly higher than those in control group (P＜0.05). In case group, the expression level of AIM2 inflammasome was statistically associated with the degree of cancer cell differentiation, FIGO stage, lymph node metastasis and high-risk HPV infection (P＜0.05),and the expression level of GSDMD was related to the degree of cancer cell differentiation, FIGO stage and vascular infiltration (P＜0.05). Conclusion The expression levels of AIM2 inflammasome and GSDMD in cervical squamous cell carcinoma tissues are significantly higher than those in normal cervical tissues and are closely related to the clinical and pathological features of cervical squamous cell carcinoma.
