中文摘要: 目的 研究KIAA1324基因与人胰腺癌细胞增殖、迁移及侵袭的关系,探索胰腺癌发生侵袭、转移的可能机制。方法培养人胰腺癌细胞系PANC-1细胞,构建KIAA1324过表达质粒并转染PANC-1细胞,使KIAA1324基因过表达并通过qPCR实验验证靶基因的表达水平;实验分3组,即空白对照组(CON组)、阴性对照组(NC-OE组)、基因过表达组(KIAA1324-OE组),Celigo及CCK-8法检测细胞增殖水平,划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力,比较CON组、NC-OE组及KIAA1324-OE组间在细胞增殖、迁移和侵袭能力方面的差异。结果 KIAA1324-OE组细胞增殖能力、迁移能力、侵袭能力高于CON组和NC-OE组,差异有统计学意义 (P<0.05)。结论 KIAA1324基因过表达可增强胰腺癌细胞系PANC-1细胞的增殖、迁移和侵袭能力。

Abstract:Objective To study the relationship between KIAA1324 gene and proliferation, migration and invasion in human pancreatic cancer cell, and to explore the possible mechanism of invasion and metastasis in pancreatic cancer. Methods KIAA1324 gene was over-expressed by culturing human pancreatic cancer cells called line PANC-1 cells, building KIAA1324 over-expressive plasmids and transfecting PANC-1 cells, and then target gene over-expressed was verified by qPCR experiments. The experiment was divided into 3 groups, blank control group (CON group), negative control group (NC-OE group) and gene over-expression group (KIAA1324-OE group). Cell proliferation level was detected by Celigo and CCK-8 method, cell migration ability was detected by scratch experiment, cell invasion ability was detected by Transwell experiment, and then the differences in cell proliferation, migration and invasiveness among three groups were compared by the above methods. Results The cell proliferation ability, migration ability and invasion ability in KIAA1324-OE group were higher than those in CON group and NC-OE group, the difference was statistically significant (P<0.05).Conclusions Over-expression of KIAA1324 gene can increase the proliferation, migration and invasion ability of PANC-1 cells in pancreatic cancer.
