中文摘要: 目的 研究姜黄素对体外高糖诱导的人视网膜血管内皮细胞(HRCECs)增殖、迁移和管腔形成能力的影响。 方法 将HRCECs分为5组进行培养:正常对照组(正常糖浓度的DMEM培养基下培养)、高糖对照组(高糖DMEM培养基下培养)、高糖+不同浓度姜黄素3个组(高糖DMEM培养基中分别加入20、40、80 μmol/L姜黄素),采用CCK-8法检测姜黄素干预12、24、48 h后各组细胞的增殖能力,采用Transwell法和Matrigel法分别检测姜黄素干预24 h后各组细胞的垂直迁移和管腔形成能力。 结果 (1)细胞增殖:与正常对照组比较,高糖对照组细胞增殖能力明显增强(P<0.01),各浓度姜黄素组作用不同时间(12、24、48 h)后与高糖对照组比较,细胞增殖抑制率明显上升(P<0.01),且呈浓度与时间依赖性,各实验组间两两比较差异均有统计学意义(P<0.01)。(2)细胞迁移:与正常对照组相比,高糖对照组细胞垂直迁移率显著增高(P<0.01);与高糖对照组相比,各浓度姜黄素组细胞垂直迁移率均显著降低(P<0.01),且随着药物浓度增加迁移率逐渐降低。(3)细胞成管:与正常对照组相比,高糖对照组细胞总管长显著增加(P<0.01);与高糖对照组相比,各浓度姜黄素组细胞总管长均显著减少(P<0.01),且随着药物浓度的增加成管率逐渐降低。 结论 姜黄素可通过抑制高糖诱导的HRCECs增殖、迁移和成管,从而抑制糖尿病视网膜病变病理性新生血管形成。
Abstract:Objective To study the effects of curcumin on the proliferation, migration and lumen formation of human retinal capillary endothelial cells (HRCECs) induced by high-sugar in vitro. Methods HRCECs were divided into five groups for culture: normal control group (cultured in DMEM medium with normal sugar concentration), high-sugar control group (cultured in high-sugar DMEM medium), three curcumin groups with different concentrations (20, 40 and 80 μmol/L curcumin was added to high-sugar DMEM medium respectively). CCK-8 method was used to detect the proliferation of cells in each group after curcumin intervention for 12, 24 and 48 h. Transwell method and Matrigel method were used to detect the vertical migration and tube formation of cells in each group after 24 hours of curcumin intervention. Results Compared with the normal control group, the cell proliferation ability of high-sugar control group was significantly increased (P<0.01). Compared with the high-sugar control group, the inhibition rate of cell proliferation in each concentration of curcumin group was significantly increased(P<0.01) in a concentration and time-dependent manner. Compared with the normal control group, the vertical migration rate of cells in the high-sugar control group was significantly increased (P<0.01). Compared with the high-sugar control group, the vertical migration rate of cells and the length of cell common duct in each concentration of curcumin group decreased significantly (P<0.01), and the migration rate and the length of cell common duct decreased gradually with the increase of drug concentration. Conclusion Curcumin can inhibit diabetic retinopathy pathological angiogenesis by inhibiting the proliferation, migration and tube formation of HRCECs induced by high-sugar.
