中文摘要: 目的 探讨芦荟大黄素对胃癌SGC-7901细胞钙联蛋白(Calnexin)/钙网蛋白(Calreticulin)循环及内质网应激(ERS)所致凋亡的影响。方法 将胃癌SGC-7901细胞分为对照组(灭活胎牛血清的MEM培养基培养)、低剂量组(SGC-7901细胞+10 μmol/L芦荟大黄素培养30 min)、中剂量组(SGC-7901细胞+30 μmol/L芦荟大黄素培养30 min)和高剂量组(SGC-7901细胞+50 μmol/L芦荟大黄素培养30 min)。细胞计数试剂盒8(CCK-8)法检测各组细胞培养24、48、72 h的细胞增殖率;脱氧核苷酸末端转移酶(TdT)介导的核苷酸(dUTP)缺口末端标记技术(TUNEL)检测各组细胞凋亡情况;实时荧光定量PCR检测各组细胞中Calnexin、Calreticulin及葡萄糖调节蛋白(GRP)78、肌醇需求酶(IRE)1、CCAAT增强子结合蛋白同源蛋白(CHOP) mRNA的变化;蛋白免疫印迹法检测各组细胞Calnexin、Calreticulin、GRP78、IRE1、CHOP的表达。结果 (1)与对照组相比,低剂量组细胞增殖率、凋亡率未见明显变化(P>0.05),中、高剂量组细胞增殖率降低、凋亡指数升高(P<0.05);(2)与对照组相比,低剂量组细胞Calnexin、Calreticulin mRNA和蛋白表达降低,而GRP78、IRE1、CHOP蛋白及mRNA表达无明显变化(P>0.05),中、高剂量组细胞Calnexin、Calreticulin蛋白及mRNA表达水平降低,GRP78、IRE1、CHOP蛋白及mRNA表达增高(P<0.05)。结论 芦荟大黄素可能通过影响SGC-7901细胞Calnexin/Calreticulin循环激活GRP78/IRE1/CHOP信号通路,加重胃癌细胞ERS,引发细胞凋亡,降低细胞增殖率。

Abstract:Objective To investigate the effects of aloe-emodin on apoptosis of gastric cancer SGC-7901 cells induced by Calnexin/Calreticulin circulating and endoplasmic reticulum stress (ERS). Methods The gastric cancer SGC-7901 cells were respectively cultured in MEM medium of inactivated fetal bovine serum (control group), 10 μmol/L aloe-emodin (low dose group), 30 μmol/L aloe-emodin (middle dose group) and 50 μmol/L aloe-emodin (high dose group) for 30 minutes. Cell Counting Kit-8 (CCK-8) was used to detect cell proliferation rates after cell culture for 24 hours, 48 hours and 72 hours. Transferase-mediated deoxyuridine triphosphate-biotin nick end labeling (TUNEL) was used to detect cell apoptosis. Real-time quantitative polymerase chain reaction assay (qRT-PCR) was used to detect the expressions of calnexin, calreticulin, glucose regulated protein 78 (GRP78), inositol-requiring enzyme 1 (IRE1) and CCAAT enhancer binding protein homologous protein (CHOP) mRNAs. Western blotting method was used to detect the expressions of calnexin, calreticulin, GRP78, IRE1and CHOP proteins. Results Compared with control group, cell proliferation rate and cell apoptosis index in low dose group remained unchanged (all P>0.05), and they increased significantly in middle dose group and high dose group(all P<0.05). Compared with control group, the expression levels of calnexin, calreticulin, GRP78, IRE1, CHOP proteins and mRNAs in low dose group remained unchanged (all P>0.05), while the expression levels of calnexin and calreticulin proteins and mRNAs decreased significantly, GRP78, IRE1, CHOP proteins and mRNAs expression levels increased significantly in middle dose group and high dose group(all P<0.05). Conclusion Aloe-emodin can induce apoptosis and reduce cell proliferation rate possibly through influencing the Calnexin/Calreticulin cycle of SGC-7901 cells, activating GRP78/IRE1/CHOP signaling pathway and aggravating ERS of gastric cancer cells.
