平喘止咳方对慢性阻塞性肺疾病模型大鼠γ-GCS、TIMP-1表达及气道重塑的影响

中文摘要: 目的 探讨平喘止咳方对慢性阻塞性肺疾病(COPD)模型大鼠γ-谷氨酰半胱氨酸合成酶(γ-GCS)、金属蛋白酶组织抑制因子-1(TIMP-1)表达及气道重塑的影响。方法 选取8周龄雄性SD大鼠30只随机分为对照组、模型组和平喘止咳方组,每组10只。对照组正常饲养,模型组和平喘止咳方组进行烟熏联合脂多糖法构建COPD大鼠模型,模型构建成功后,平喘止咳方组给予5 ml/kg平喘止咳方灌胃,对照组和模型组给予5 ml/kg生理盐水灌胃,每日1次,共给药14 d。比较各组肺功能、肺组织病理变化、气道重塑情况、肺组织γ-GCS、TIMP-1表达情况,并分析肺组织γ-GCS、TIMP-1表达与气道重塑的相关性。结果 各组大鼠第0.1秒用力呼气量/用力肺活量(FEV0.1/FVC)、呼气流量峰值(PEF)比较,差异有统计学意义(模型组<平喘止咳方组<对照组,P<0.05)。三组肺组织损伤评分比较差异有统计学意义(模型组>平喘止咳方组>对照组,P<0.01)。各组大鼠气管壁厚度、气管平滑肌厚度比较,差异有统计学意义(模型组>平喘止咳方组>对照组,P<0.01)。各组大鼠肺组织γ-GCS、TIMP-1 mRNA表达水平比较,差异有统计学意义(模型组>平喘止咳方组>对照组,P<0.01)。γ-GCS、TIMP-1蛋白表达与气管壁厚度分别呈正相关(r=0.453,P=0.012;r=0.540,P=0.002)。结论 平喘止咳方能有效提高COPD大鼠肺功能,减轻肺部病理变化,改善气道重塑,其机制可能与下调γ-GCS、TIMP-1表达水平有关。
Abstract:ObjectiveTo investigate the effect of Pingchuan Zhike Decoction on the expression of γ-glutamylcysteine synthase (γ-GCS),tissue inhibitor of metalloproteinase-1 (TIMP-1) and airway remodeling in rats with COPD.MethodsA total of 30 8-week-old male SD rats were selected and randomly divided into control group,model group and Pingchuan Zhike Decoction group (n=10,each).The control group was fed normally,the fumigating and lipopolysaccharide method was used to construct COPD rat model in the model group and the Pingchuan Zhike Decoction group.After the model was successfully constructed,the Pingchuan Zhike Decoction group was given Pingchuan Zhike Decoction by gavage (5 ml/kg),and the control group and model group were given normal saline by gavage(5 ml/kg),one time per day.The lung function,pathological changes of lung tissue,airway remodeling,expression of γ-GCS and TIMP-1 in lung tissue were compared,and the correlation between the expression of γ-GCS and TIMP-1 and airway remodeling were analyzed.ResultsThere were significant differences in FEV0.1/FVC,PEF between three groups (model groupPingchuan Zhike Decoction group>control group,P<0.01).The expressions of γ-GCS and TIMP-1 protein were positively correlated with tracheal wall thickness(r=0.453,P=0.012;r=0.540,P=0.002).ConclusionsPingchuan Zhike Decoction could improve the lung function,alleviate the pathological changes of lung and improve the airway remodeling in COPD rats.The mechanism may be related to the down-regulation of the expression levels of γ-GCS and TIMP-1.
